Engineering of Ancestors as a Tool to Elucidate Structure, Mechanism, and Specificity of Extant Terpene Cyclase
An article from Mistra SafeChem is now published in JACS, Journal of the American Chemical Society. One of the authors is Per-Olof Syrén, KTH, who explains the findings:
"Our work resolves current bottlenecks associated with the implementation of enzymes in green chemical manufacturing, namely stability, specificity and activity of biocatalyst. We show that reconstructed engineered ancestors enable studying extant biosynthetic enzymes that are challenging to crystallize to elucidate their mechanisms and redesign catalytic versatility. We envision that this method will find broad implementation in next-generation chemical production."
